Mechanistic Studies Lead to Dramatically Improved Reaction Conditions for the Cu-Catalyzed Asymmetric Hydroamination of Olefins

چکیده
Enantioselective copper(I) hydride (CuH)-catalyzed hydroamination has undergone significant development over the past several years. To gain a general understanding of the factors governing these reactions, kinetic and spectroscopic studies were performed on the CuH-catalyzed hydroamination of styrene. Reaction profile analysis, rate order assessment, and Hammett studies indicate that the turnover-limiting step is regeneration of the CuH catalyst by reaction with a silane, with a phosphine-ligated copper(I) benzoate as the catalyst resting state. Spectroscopic, electrospray ionization mass spectrometry, and nonlinear effect studies are consistent with a monomeric active catalyst. With this insight, targeted reagent optimization led to the development of an optimized protocol with an operationally simple setup (ligated copper(II) precatalyst, open to air) and short reaction times (<30 min). This improved protocol is amenable to a diverse range of alkene and alkyne substrate classes.

Catalytic Diamination of Olefins via N–N Bond Activation
CONSPECTUS: Vicinal diamines are important structural motifs present in various biologically and chemically significant molecules. Direct diamination of olefins provides an effective approach to this class of compounds. Unlike well-established oxidation processes such as epoxidation, dihydroxylation, and aminohydroxylation, direct diamination of olefins had remained a long-standing challenge an...
متن کاملMerging synthesis and enantioselective functionalization of indoles by a gold-catalyzed asymmetric cascade reaction.
Significance: The authors developed a gold-catalyzed hydroamination followed by an asymmetric nucleophilic allylic substitution cascade to access oxazino[4,3-a]indoles in good yields and enantioselectivities. Owing to pharmacologically active indole alkaloids, this rapid synthesis and asymmetric decoration of the polycyclic indolyl core is highly important. Comment: Amazingly, the stereodiffere...
متن کاملStudies on enantioselective allylic oxidation of olefins using peresters catalyzed by Cu(I)-complexes of chiral pybox ligands.
Enantioselective allylic oxidation of olefins with various peresters, using a catalytic amount of Cu(I)-pybox complex, can be tuned to achieve high asymmetric induction (up to 98% ee) by choosing a unique combination of a ligand and a perester at room temperature. The asymmetric induction in the reaction strongly depends on the nature of the substituents attached to the aryl ring of peresters. ...
متن کاملDevelopment of catalysts for the hydroamination of olefins*
Studies on the development of palladium, nickel, and rhodium catalysts for the hydroamination of dienes and vinylarenes are described. Enantioselective catalysts based on palladium have been developed for the addition of arylamines to dienes and for Markovnikov addition of arylamines to vinylarenes. In addition, nickel catalysts for the addition of aliphatic amines to dienes have been developed...
متن کاملIridium-catalyzed intermolecular hydroamination of unactivated aliphatic alkenes with amides and sulfonamides.
The intermolecular addition of N-H bonds to unactivated alkenes remains a challenging, but desirable, strategy for the synthesis of N-alkylamines. We report the intermolecular amination of unactivated α-olefins and bicycloalkenes with arylamides and sulfonamides to generate synthetically useful protected amine products in high yield. Mechanistic studies on this rare catalytic reaction revealed ...
